Overcoming Matrix Effects in the Analysis of Lipophilic Compounds

Highly lipophilic compounds often co-elute with structurally similar compounds, presenting significant analytical challenges. In the absence of a stable-isotope-labeled (SIL) internal standard, these challenges frequently lead to matrix effects during method validation. Furthermore, when such compounds exhibit instability, maintaining sample integrity throughout the preparation process demands strict conditions and meticulous handling.

At the 10th EBF Young Scientist Symposium, we highlighted the complexities of analysing Fucoxanthin derivatives and demonstrated how innovative approaches, such as 2D Chromatography, can overcome these obstacles. These advanced techniques not only enable the accurate analysis of lipophilic compounds but also support robust method validation in compliance with ICH M10 guidelines.
